چکیده
Nuclear factor-κB (NF-κB) is important in inflammatory skin diseases, including contact dermatitis (Th1 inflammation), atopic (Th2 and psoriasis (Th17 inflammation). In fact, molecularly targeted drugs against tumor necrosis factor (TNF), which both upstream downstream of NF-κB have been demonstrated to be effective diseases such as psoriasis. The linear ubiquitination by ubiquitin chain assembly complex (LUBAC), consists HOIL-1L, HOIP SHARPIN, activating IKKβ, essential for TNF/NF-κB signaling. It has that activates TNF/IKKβ/NF-κB signaling, while HOIL-1L inhibits it. this study, investigate the importance signaling we used mice lacking RING1 domain, active site (HOIL-1LΔRING1 mice). these mice, suppressed enhanced. HOIL-1LΔRING1 did not differ from wild-type (WT) control either a Dinitrofluorobenzene-induced model or an MC903-induced model. On other hand, imiquimod-induced model, showed more severe clinical manifestations than controls. To examine whether radio-resistant cells immune cells, generated bone marrow (BM) chimeric mice. When BM WT were transplanted was worse group with when there no difference dermatitis. These results indicate pathogenesis inhibitors targeting may new therapeutic target drug discovery

The linear ubiquitin chain assembly complex regulates TRAIL‐induced gene activation and cell death
The linear ubiquitin chain assembly complex (LUBAC) is the only known E3 ubiquitin ligase which catalyses the generation of linear ubiquitin linkages de novo LUBAC is a crucial component of various immune receptor signalling pathways. Here, we show that LUBAC forms part of the TRAIL-R-associated complex I as well as of the cytoplasmic TRAIL-induced complex II In both of these complexes, HOIP li...

UNLABELLED Constitutive activation of NF-κB is a hallmark of the activated B cell-like (ABC) subtype of diffuse large B-cell lymphoma (DLBCL), owing to upstream signals from the B-cell receptor (BCR) and MYD88 pathways. The linear polyubiquitin chain assembly complex (LUBAC) attaches linear polyubiquitin chains to IκB kinase-γ, a necessary event in some pathways that engage NF-κB. Two germline ...
متن کاملActivation of nuclear factor-kappa B by linear ubiquitin chain assembly complex contributes to lung metastasis of osteosarcoma cells.
NF-κB is involved in the metastasis of malignant cells. We have shown that NF-κB activation is involved in the pulmonary metastasis of LM8 cells, a highly metastatic subclone of Dunn murine osteosarcoma cells. Recently, it was determined that a newly identified type of polyubiquitin chain, a linear polyubiquitin chain, which is specifically genera...
متن کاملA non-canonical UBA-UBL interaction forms the linear-ubiquitin-chain assembly complex.
HOIL-1L and its binding partner HOIP are essential components of the E3-ligase complex that generates linear ubiquitin (Ub) chains, which are critical regulators of NF-κB activation. Using crystallographic and mutational approaches, we characterize the unexpected structural basis for the specific interaction between the Ub-like domain (UBL) of HOIL-1L and the Ub-associated domain (UBA) of HOIP....
متن کاملSpecific recognition of linear ubiquitin chains by the Npl4 zinc finger (NZF) domain of the HOIL-1L subunit of the linear ubiquitin chain assembly complex.
The linear ubiquitin chain assembly complex (LUBAC) is a key nuclear factor-κB (NF-κB) pathway component that produces linear polyubiquitin chains. The HOIL-1L subunit of LUBAC has been shown to bind linear chains; however, detailed structural and functional analyses on the binding between LUBAC and linear chains have not been performed. In this study, we found that the Npl4 zinc finger (NZF) d...
